De Sanders signed her first contract with Ajax in the summer of 2017. She made her official debut on November 3 of that same year in a 2–0 victory over Excelsior. In total, she made 147 appearances for the Ajax Women across eight seasons.
The away match against Excelsior on March 10, 2023 was a special occasion for the defender. De Sanders played her 100th match for Ajax that day, earning her a place in the 'Club of 100'.

De Sanders ended her very first Eredivisie season by winning the league title with Ajax. That same year, she also lifted her first KNVB Cup after a 3–1 victory over PSV. In the 2022/2023 season, De Sanders became Dutch champion for the second time.
In the 2018/2019 season, Ajax beat PEC Zwolle 2–1, allowing De Sanders to lift the KNVB Cup for the second year in a row. In 2021/2022, Ajax once again faced PSV in the final, claiming a 2–1 win. Her most recent trophy came this past season, when Ajax defeated Fortuna Sittard in the KNVB Cup final.
